黄瓜23对高多态性SSR标记的筛选与评价

摘要: 目前在黄瓜种质研究中仍然缺乏可用的引物。利用黄瓜基因组连锁遗传图谱开发的995对备选SSR引物,挑出在7条染色体上均匀分布且能扩增出清晰稳定单一条带的多态性引物23对,并利用这些引物对黄瓜种质遗传多样性进行了探讨。通过来自不同国家和地区的30份代表性黄瓜种质资源进行SSR扩增分析,从中能够得到153条多态性条带。不同引物分析得到的种质遗传多样性差异较大,供试种质间平均期望杂合度为0.57,平均PIC值(多态性信息量)为0.60。23对引物可以清晰检测出亚洲、欧美和印度群体遗传多样性,较客观地反映了群体结构和地域性差异等信息。

Abstract: At present,useful primers in the study of cucumber germplasm are scarce,in this study,23 SSR primers are selected from the 995 primers of cucumber genome linkage map,the 23 SSR primers distribute on the linkage map uniformly and can amplify clear,stable and single bands,using these markers,the genetic diversity of 30 cucumber(Cucumis sativus L.)accessions from different countries or regions was detected,and 153 polymorphism bands were got. Genetic diversity differs among 23 primers. The expected heterozygosity of detected resources is 0.57,and the average of PIC(Polymorphism information content)value is 0.60. The results distinctly prove the genetic diversity among Asian,European and India resources by 23 primers and objectively reflect information including population structure and regional differences.
