安全标记基因pmi在果树转基因研究中的应用进展

摘要: 关于木本植物转基因成功的报道很多,但木本果树的转化效率较低。甘露糖/PMI筛选体系以pmi为安全标记基因,实现对转化细胞安全、高效地正向选择。从甘露糖/PMI筛选体系的作用原理,影响其筛选效率的主要因素,与其它筛选体系的对比优势(高效、安全、易于检测)等几个方面综述了这一筛选体系在木本果树转基因研究上的应用现状及其发展前景。

Abstract: Despite the production of fertile transgenic plants through transformation mediated by Agrobacterium tumefaciens,transformation efficiency is still low in fruit trees. The mannose selection system employs the phosphomannose isomerase(PMI)gene as selectable gene and the transgenic PMI-expressing cells have acquired the ability to convert mannose-6-phosphate to fructose-6-phosphate. Compare to antibiotic selection system,it has advantages such as high selective efficiency,safety for the environment and easy to test. It is also affected by some important factors. Here we talk about the application of this system on fruit trees transformation and its perspective based on the existing literature data. The advantages of this select system,some important factors affecting the transformation efficiency are discussed as well as the approaches in fruit tree genetic transformation using this PMI/mannose system.
