菜豆抗炭疽病基因SCAR标记在品种抗性鉴定中的应用

Application of Common Bean Anthracnose Resistance Gene SCAR Markers in Snap Bean Disease Resistance Identification

摘要: 利用5个来自于普通菜豆抗炭疽病基因SCAR 标记(SCA1000、SH181100、SAB3400、SB12350和SCF101072)引物组,对143份荚用菜豆资源进行DNA水平的鉴定。同时使用苗期接种方法,验证了143份资源的抗病性。结果表明:共有61份资源检测到SCAR标记。在这些资源中,有40份具有SCA1000标记,40份具有SCF101072标记,19份兼有SCA1000和SCF101072标记,其余的82份未检测到SCAR标记。这143份资源中均未检测到SAB3400、SB12350和SH181100扩增带。群体抗性鉴定结果显示:143份资源中,抗病的有16份。其中含有2个SCAR标记,群体抗性结果表现抗病的有5份,含有1个SCAR标记,并且群体抗性结果表现抗病的有3份。通过本研究明确了:荚用菜豆中Co-2和Co-10基因含量比较丰富,除扁圆荚类型紫荚菜豆中Co-2基因含量多于Co-10基因外,这两个抗病基因在其它类型的菜豆中分布比较均匀。在油豆类型和扁圆荚类型绿荚菜豆资源中,同时含有此两个抗病基因标记的较多。在油豆类型资源中,含有两个抗病标记,并且群体抗性表现抗病的最多。

Abstract: Five SCAR primer combinations(SCA1000,SH181100,SAB3400,SB12350 and SCF101072)of common bean anthracnose resistance gene were used to check the genome DNA of 143 snap bean germplasms. Meanwhile,their anthracnose resistance was identified by seedling inoculation method. The results indicated that the SCAR markers were found in 61 out of 143 germplasms,40 germplasms were detected having marker SCA1000,40 germplasms were detected having marker SCF101072,19 germplasms were detected having both marker SCA1000 and SCF101072. SCAR markers,SAB3400,SB12350 and SH181100,were not detected in all germplasms. Population resistance results revealed that 16 germplasms out of 143 tested germplasms were disease resistant. Among them,5 germplasms possessed two SCAR markers and disease resistance,3 germplasms possessed only one of the two SCAR markers with disease resistance. The results also showed that the content of Co-2 and Co-10 are very rich and distribute evenly in snap bean germplasms,except that the Co-2 content of is higher than Co-10 in purple bean pod. There are more resistance gene in green pods and You Dou. Numbers of owning both resistance gene and disease resistance are the most in You Dou.
