黄瓜ARF家族序列特征及部分成员在果实发育早期的表达分析

摘要: 采用生物信息学方法获取黄瓜ARF家族信息,并对其进行染色体、序列相似性以及系统进化树分析,同时选取与单性结实基因亲缘关系较近为主的9个ARF基因,以单性结实自交系‘6401’和非单性结实自交系‘6429’为试材,在花后0、2和4 d取‘6401’和‘6429’授粉和未授粉两种处理的果实,利用半定量RT-PCR进行表达分析。结果显示:18个ARF基因遍布于黄瓜的7条染色体上,氨基酸长度在172 ~ 1 102残基之间,相似性在4.41% ~ 62.12%的范围内,基因重复、序列差异和基因缺失是黄瓜ARF家族的主要进化模式;基因Csa021954和Csa012805在果实中受授粉诱导而表达;Csa019265、Csa009209和Csa009210在所有样品中都有较高的转录本;基因Csa012237和Csa015176在果实早期发育中表现出衰减趋势;Csa019264和Csa010564只在发育的果实中表达且表达水平较高,推测这两个基因的高水平表达可能对果实膨大起促进作用。

Abstract: Eighteen ARF family members were identified from Cucumber Genome Datebase using bioinformatics methods. They are distributed on 7 chromosomes,and their evolutionary patterns are the gene duplication,sequence differences and gene deletion. With RT-PCR technique,parthenocarpic line ‘6401’and non-parthenocarpic line‘6429’were used to test the expression patterns of 9 ARF genes that were selected by their homology with genes related parthenocarpy. The results of Csa021954 and Csa012805 indicated that they were only induced in the fruits from pollination. However,Csa019265,Csa009209 and Csa009210 have more transcripts in all samples. At the same time Csa012237 and Csa015176 showed significant reduction in the early development fruits of cucumber. Furthermore,the    expression of Csa019264 and Csa010564 only are in the development fruits and higher levels. Therefore,it was speculated that the these genes may play an important role in promoting fruit development of cucumber.
