月季‘萨蔓莎’愈伤组织的诱导及植株再生

Callus Induction and Plant Regeneration of Rosa hybrida ‘Samantha’

摘要: 以试管苗的小叶为外植体, 探讨了月季品种‘萨蔓莎’愈伤组织诱导及植株再生的方法。结果表明: 高浓度的生长素能诱导小叶产生愈伤组织, 经2,4-D 7.0~10.0 mg/L光下诱导愈伤15 d的小叶外植体, 在含TDZ 1.5 mg/L的MS培养基上光下培养约20 d后有白色假珠芽形成, 此芽暗培养在含NAA、ZT和GA3 的培养基上产生新的假珠芽和新的愈伤组织, 通过持续选择继代生活力高的组织, 约1年后获得可长期继代的愈伤组织, 到目前为止已保存了16个月。此愈伤组织在TDZ 1.0 mg/L、NAA 0.01 mg/L和GA3 0.1 mg/L的诱导下30 d内分化出不定芽, 在含BA 2.0 mg/L和NAA 0.01 mg/L的培养基上不定芽伸长形成正常植株。

Abstract: White pseudobulbils were initiated from in vitro grown leaf explants of Rosa hybrida ‘Samantha’following an induction period of 15 d on MS basal medium supp lemented with 2,4-D 7.0 - 10.0 mg/L and a subculture of 20 d on MS medium with TDZ 1.5 mg/L under light. The pseudobulbils were subcultured
on medium with NAA, ZT and GA3 , and during the process, new pseudobulbils and new calluswere formed. After a year of continuous selection for fresh and nodular callus, callus that could be continuously subcultured was obtained, and has been maintained for 16 months till now. Adventitious buds could be induced from the callus onMS medium with TDZ 1.0 mg/L, NAA 0.01 mg/L and GA3 0.1 mg/L and elongated into shoots on MS medium with BA 2.0 mg/L and NAA 0.01 mg/L.
