干旱胁迫下柑橘叶片基因表达谱的cDNA-AFLP 分析

Gene Expression Profiling in Response to Drought Stress in Citrus Leaves by cDNA-AFLP

摘要: 为分离和鉴定柑橘干旱胁迫应答基因,应用cDNA-AFLP分析技术,以盆栽‘天草’橘橙枳砧嫁接苗为试材,对正常灌水和不同干旱胁迫处理下叶片的基因表达谱进行分析。利用81对引物组合进行扩增,共筛选获得113条差异表达转录衍生片段(TDF),其中上调表达71条(63%),下调表达29条(26%),其余13条(11%)为瞬时表达。选取70个差异表达TDF进行克隆测序,共得到62个有效序列,BLAST结果表明:33个TDF与已知功能基因具较高同源性,功能涉及信号转导、能量代谢、离子通道和蛋白合成等,18个TDF与功能未知基因或假定基因同源性较高,其余11个TDF无同源序列,可能为新的未知功能基因。

Abstract: Transcripts involved in drought stress in‘Amakusa’tangor(Citrus reticulata × C. sinensis)were identified by cDNA-AFLP gene expression profiling in leaves under normal watering and water deficit condition. A total of 113 differentially expressed transcript-derived fragments(TDFs)were identified by selective PCR amplification using 81 primer combinations. The TDFs included 71(63%)upregulated,29(26%)downregualted,and 13(11%)transiently expressed genes. Seventy TDFs were recovered,sequenced,and functionally annotated using sequence homology to GenBank entries. Out of the 70 TDFs,33 are homolog to genes with a known function,18 are similar to genes with unknown function,and 11 have not sequence homology to GenBank entries. The function of homologous genes included signal transduction,energy metabolism,ion channel,and protein synthesis.
