Obtaining of Ponkan (Citrus reticulata Blanco ) Tetraploid by Culturing Embryos of Mature Seeds

摘要: 通过组培方法从374粒 柑种子培养中获得449株小苗。利用流式细胞仪进行染色体倍性分析, 发现其中7株小苗为四倍体植株。采用13对SSR引物分析表明这7株四倍体植株均来自珠心细胞, 为同源四倍体。这些四倍体的获得为下一步培育三倍体无核品种奠定了基础。

Abstract: Seedless or few seed trait is one of the goals of Ponkan breeding. By culturing the embryos of mature seeds, a total of 449 polyembryonic seedlings from 374 seeds were obtained; and seven of them were verified to be tetrap loid by flow cytometry. SSR (simple sequence repeat) analyseswith 13 p rimers indicated
that these tetraploid seedlingswere all autotetrap loid from the nucellar cells. These tetraploid seedlings will be of great value for creating seedless Ponkan through sexual hybridization.
