辣椒子叶高效植株再生体系的建立

摘要: 以双丰等17 个辣椒品种为试材, 探讨了不同基因型、激素组合、有机成分及AgNO3 等因素对子叶再生的影响。观察到DJ 添加物(辣椒幼苗茎叶提取汁液∶卡那霉素= 500∶1) 对不定芽的分化及伸长有明显的促进作用, 比对照分别提高10. 0 %~11. 1 %和90. 5 %~133. 3 %。筛选出高效芽分化培养基为MB+ IAA 1. 0 mg/L + 62BA 5. 0 mg/L + DJ 5 000. 0 mg/L + AgNO3 10. 0 mg/L , 17 个品种平均芽分化率达92. 3 %;高效芽伸长培养基为MB + IAA 1. 0 mg/L + 62BA 5. 0 mg/L + DJ 5 000. 0 mg/ L + AgNO3 10. 0 mg/ L + GA3 2. 0 mg/ L , 17 个品种平均芽伸长率达57. 8 %; 高效生根诱导培养基为MS + IAA 0. 2 mg/ L + NAA 0. 1 mg/ L ,生根率达90 % , 建立了辣椒子叶高效植株再生体系。

Abstract: The report describes the development of a high efficient in vitro regeneration procedure from cotyledon explants of 17 sweet/ chilli pepper cultivars (Shuangfeng et al) . The effect of genotype , hormone , organic nutrient component and AgNO3 et al on the tissue culture from pepper cotyledon explants was investigated. Adding DJ(juice of peppr stem and leaf + kanamycin = 500∶1) organic nutrient component can improved the shoot buds differentiation and elongation rate. The best media for in vitro regeneration from cotyledon explants of pepper were discovered : the shoot buds induction medium: MB + IAA 1. 0 mg/ L + 62BA 5. 0 mg/ L + DJ 5. 0 g/ L + AgNO3 10. 0
mg/ L ; the shoot buds elongation medium: MB + IAA 1. 0 mg/ L + 62BA 5. 0 mg/ L + DJ 5. 0 g/ L + AgNO3 10. 0 mg/ L + GA32. 0 mg/ L ; the rooting medium: MS + IAA 1. 0 mg/ L + NAA 0. 1 mg/ L.
