Isolation of Cold Stress Genes through cDNA-AFLP from Volvariella volvacea

摘要: 采用cDNA-AFLP技术筛选获得了5个草菇冷诱导表达基因片段VC1、VC2、VC3、VC4 和
VC5。测序结果表明, 5个DNA片段大小分别为247 bp、219 bp、172 bp、350 bp和171 bp。同源性BLAST搜索结果显示, VC1、VC2、VC3、VC4和VC5片段目前都没有同源的核酸序列; VC1片段翻译的蛋白质序列与粗糙脉孢菌(Neurospora crassa) 的假拟蛋白具有一定的同源性, VC2片段翻译的蛋白质序列与水稻的AMP脱氨酶具有一定的同源性, VC3片段翻译的蛋白质序列与Danio rerio的糖原合酶激酶3α具有较高的同源性, VC4片段翻译的蛋白质序列与Leuconostocm esenteroides的假拟蛋白有一定的同源性, VC5片段翻译的蛋白质序列没有搜索到同源序列。

Abstract: Five cold stress gene fragments were isolated through cDNA-AFLP from Volvariella volvacea and were designated as VC1, VC2, VC3, VC4 and VC5 respectively. DNA sequence results indicated that these five DNA fragments had 247 bp, 219 bp, 172 bp, 350 bp and 171 bp respectively. BLAST search results showed that VC1, VC2, VC3, VC4 and VC5 had no significant homologous nucleotide sequences found in databases. Postulated p rotein sequences of VC1, VC2 were homologous with the hypothetical protein of
Neurospora crassa and the putative AMP deaminase of Oryza sativa, espectively. Postulated protein sequences of VC3, VC4 were homologouswith the glycogen synthase kinase three alpha in Danio rerio and the hypothetical protein in Leuconotocm esenteroides respectively. But postulated protein sequence of VC5 had not found the significant homologous p rotein sequence in the databases yet.
