菠萝愈伤组织中体细胞胚起源过程的组织细胞学观察

摘要: 以菠萝(Ananas comosus)‘神湾’品种为材料,采用石蜡切片法对其愈伤组织中球形胚形成过程进行了组织细胞学观察。结果表明,非胚性愈伤组织转入到MS + 4.0 mg · L-1 2,4-D + 0.5 mg · L-1 6-BA培养基培养后,愈伤组织中先出现一些胚性细胞;然后单个胚性细胞分裂形成一个由多细胞组成的原胚。原胚细胞分生能力强,与周围的母体组织之间形态差异显著,界限明显,其下部形成一个类似胚柄的结构。随着原胚不断生长,其形状由长椭圆形发展成倒卵形,最后逐渐形成表面较光滑、外观近球形的球形胚,并在‘胚柄’基部与母体组织连接处形成离层。与此同时,球形胚内出现细胞分化。菠萝体细胞胚发生具有内、外两种起源方式,另外愈伤组织中体细胞胚发生普遍存在着不均匀、不同步和次生胚等现象。

Abstract: Histocytology study was done by paraffin section method on the globular embryogenesis in pineapple(Ananas comosus). The results showed that the non-embryonic callus were cultured on the MS medium contained 4.0 mg · L-1 2,4-D and 0.5 mg · L-1 6-BA, embryogenic cells appeared in it. Then the single embryogenic cells formed a proembryo composed of many cells through cell division. The cells of proembryo had strong division ability, and there were distinct difference in shape and obvious boundary between them/it and the surrounding tissues. A suspensor-like structure was formed on the bottom of the proembryo. Along with the constant growth of the proembryo, its shape converted from long-ellipse to obovate, at last it evolved as a globular embryo with smooth surface, and there were absciss layer in the joint of the bottom of suspensor and matrix tissue. At the same time, cell differentiation appeared in the globular embryo. Pineapple somatic embryogenesis has two original modes,endogenous origin and exogenous origin. Additionally, there were ubiquitous uneven,desynchronization and secondary embryosphenomenon during somatic embryogenesis in pineapple callus.
