洋葱T型细胞质雄性不育相关基因的结构与表达模式研究

摘要: orfA501标记与洋葱细胞质雄性不育表型之间的完全符合引起研究者对其所在基因结构的兴趣。试验通过锚定PCR方法获得了orfA501的侧翼序列,明确了其在洋葱T型细胞质雄性不育线粒体基因组中的位置;采用RT-PCR方法研究了orfA501及其相连基因的转录模式。结果表明:洋葱T型细胞质雄性不育线粒体基因组中orfA501 的5′ 端与coxⅠ基因紧密相连,二者相隔93个碱基;orfA501的3′端是由atp9、atp6、atp1以及nad4等基因片段构成的一个复杂序列;在洋葱T型细胞质雄性不育系花药中,orfA501与coxⅠ基因分别转录,且orfA501对coxⅠ的转录具有正调控的作用。

Abstract: The perfect coincidence of orfA501-marker and the phenotype of onion cytoplasmic male sterile has arose interesting in researchers. In this experiment,flanking sequences of orfA501 in mitochondrial genome of onion T-type cytoplasmic male sterility was obtained successfully by anchored PCR,and expression pattern of the chimeric gene was analysed by real time PCR. The result showed that:It was cytochrome c oxidase subunit I(coxⅠ)gene who flanked 5′ of orfA501,and a complicated sequence composed by fractions of atp9,atp6,atp1 and nad4 located in the 3′ region of orfA501. Though there were 93 bases between orfA501 and coxⅠ,they transcripted seperately in the anthors of T-type cytoplasmic male sterility,and orfA501 seemed to play an up-regulator factor for the transcription of coxⅠ.
