黄瓜ω-3脂肪酸去饱和酶基因cDNA的克隆及表达

摘要:

利用cDNA末端快速扩增技术, 从黄瓜叶片组织中获得脂肪酸去饱和酶基因(Cs-FAD),其序列长度为1 807 bp,编码445个氨基酸,推导的氨基酸序列与已经报道的ω-3脂肪酸去饱和酶基因序列同源性较高,与桃果实的同源性最高(77%)。利用RT-PCR半定量分析显示,该基因在绿色组织如茎、叶、卷须里表达量较高,在根、花、果实及种子里也有少量表达。

Abstract: The fatty acid deasturase gene (Cs-FAD), which full-length is 1 807 bp and encode 445 amino acids, was cloned from cucumber leaves by technique of rapid PCR amplification cDNA-ends. High similarity was observed between the sequence of Cs-FAD and FAD of peach ( identity, 77%).The result of RT-PCR simi-quantitative analysis showed that Cs-FAD expresses higher in green tissues, such as leaves, stems, hooks etc. And it could also be found expression in roots, flower, fruits and seeds, in a low level.
