白菜β-1,3 - 葡聚糖酶基因cDNA的克隆及序列分析

Cloning and Sequence Analysis of Beta-1,3-glucanase Gene cDNA in Brassicacampestris ssp. chinensis

摘要: 以白菜抗霜霉病品种‘雪克青’cDNA 为模板, 采用RT2PCR 技术, 获得了1032 bp 的β- 1,3
- 葡聚糖酶基因的cDNA 序列。序列分析表明, 克隆的β- 1,3 - 葡聚糖酶基因cDNA 序列编码343 个氨基酸, 与芜菁bg1 基因具有98 %的同源性, 与拟南芥bg2 基因具有84%的同源性。在GenBank 中登录号为AY395720 。

Abstract: The beta-1,3-glucanase gene cDNA sequence were cloned using RT-PCR with cDNA isolated f rom Brassica campestris ssp. chi nensis‘Xuekeqing’, which displayed resistances to downy mildew.Sequencing result indicated that cloned f ragment of beta21032glucanase gene encode a peptide containing 343amino acids. The further comparison to turnip beta-1,3-glucanase ( bg1) gene and Arabidopsis thaliana beta-1,3-glucanase ( bg2) gene showed that identity was 98 % and 84 % , respectively. As a result , the sequence was accepted and released by GenBank (accession number AY395720) .
